Lupron Depot (leuprolide acetate for depot suspension) is a prescription injectable medication administered by your doctor as a single intramuscular injection at scheduled intervals. It is FDA- approved for advanced prostate cancer in men, management of endometriosis in women, preoperative treatment of anemia caused by uterine fibroids, and central precocious puberty in children (Lupron Depot-Ped). Available in multiple strengths designed for 1-month, 3-month, 4-month, or 6-month dosing intervals.
How it works:
Your body produces sex hormone — testosterone in men, estrogen and progesterone in women — through a cascade beginning in the brain. The hypothalamus releases GnRH, which signals the pituitary gland to release LH and FSH. These hormones, in turn, stimulate the testes to produce testosterone or the ovaries to produce estrogen. Many conditions — prostate cancer, endometriosis, uterine fibroids — are fueled by these sex hormones.
Lupron Depot initially overstimulates the GnRH receptors in the pituitary gland. During the first 1–2 weeks, this overstimulation causes a temporary surge in sex hormones. However, the continuous, sustained stimulation from the depot injection then causes the pituitary’s GnRH receptors to become desensitized and shut down — a process called downregulation. Once downregulated, the pituitary stops sending signals to produce testosterone or estrogen, and hormone levels drop to near-castrate levels within 2–4 weeks. For prostate cancer, eliminating testosterone starves cancer cells. For endometriosis, lowering estrogen shrinks endometrial implants. For fibroids, reducing estrogen shrinks fibroids and reduces heavy bleeding.
Form and use:
Administered as a single intramuscular injection by your healthcare provider at scheduled intervals — every 1, 3, 4, or 6 months depending on the formulation prescribed. Strengths include 3.75 mg, 7.5 mg, 11.25 mg, 22.5 mg, 30 mg, and 45 mg. Lupron Depot-Ped is administered in pediatric dosing for central precocious puberty.
Generic availability:
As of April 2026, no generic version of Lupron Depot (the long-acting depot injectable) is available in the United States. While a generic daily leuprolide acetate injection exists (primarily used in IVF protocols) and is available for as little as $58 to $82 with coupons, this daily injection is a fundamentally different product — it does not provide the sustained, long-acting hormone suppression that Lupron Depot delivers with a single injection lasting 1–6 months.
